If I purchase the Minatomirai-Yokohama Daily Pass which allows me unlimites rides for 500 Yen, is it also valid for use on the Akai Kutsu Bus?

No, it is a train pass only:

Maybe the Minato Burari Ticket covering the subway and buses (including Akai Kutsu bus service) would be better for you, also for 500:
