Theoretically it is possible, but you won't have much time at Fuji Highland. I hear waiting times for the attractions can be long, meaning that you might not be able to try out many rides, at all.

if yes how

It is explained here:
http://www.japan-guide.com/e/e6905.html

Train timetables can be looked up here:
http://www.hyperdia.com/

The correct spelling is Kawaguchiko. There is also a "Fujikyu Highland" Station.

Bus timetables are here:

Beside the waiting times you have also to be aware about the entrance fee of Fuji Q Highland. To my opinion it's to much for a few hours visit. If possible you have the option to stay at the own Fuji Q resort (or another Hotel in the area) over night and do the other things on the next day.

To get a feeling about the waiting times, I have visited Fuji Q on a weekday outside of the peak season and had waiting times of approximately 60 Minutes at the major attractions and 0-5 Minutes at the minor attractions.

However, the second part of the train journey (Otsuki - Fuji Q Highland) offers you some great views on Mt Fuji, if the sky is clear.

We had wait times of 2-3.5hrs on the 4 major rides, and 1.5-2.5hrs for others. It was a Tuesday in September. Waste of a day pass for sure.

It is possible to travel from Kawaguchiko through the mountains to Takayama via Kofu and Matsumoto:

Kawaguchiko - (bus) - Kofu - (train) - Matsumoto - (bus) - Takayama

Here is the timetable:
http://bus.fujikyu.co.jp/line/jikokuhyo/03.html
xmRw Fujisan Station
͌Ήw Kawaguchiko Station
b{w Kofu Station

The ones with a star don't operate on weekends and public holidays. The ones with a triangle operate on weekends and holidays only.

The one way fare is 1430 yen from Kawaguchiko 1530 yen from Fujisan Station.

Alternatively you can travel by train via Otsuki, but it usually takes longer and is probably also more expensive.

Bus from Matsumoto to Takayama
