Take the JR Yamatoji Line (also known as Kansai Line) from central Osaka to Kamo Station (45-60 minutes). Direct Kamo-bound trains depart from Osaka Station except in the early mornings when they depart from Namba Station. All trains stop at Tennoji Station along the way. At Kamo Station, transfer to another train on the JR Kansai Line to Iga-Ueno Station (35 minutes) and finally change to the Iga Railway to Ueno-shi Station (5 minutes).
From Tennoji Station, the entire trip takes about 100 minutes and costs 1600 yen (1340 yen for the JR part and 260 yen for the Iga Railway part). There is generally one connection per hour. The trip is covered by the Japan Rail Pass, the JR Kansai Area Pass and the JR Kansai Wide Area Pass, except for the section of the Iga Railway.
